RNA molecules have various applications in living cells, such as 1) transmission and translation of genetic information, 2) structural support of molecular machines, 3) regulation of gene expression, silencing and catalysis. Obtaining a complete spatiotemporal distribution of RNA synthesis, processing, and transportation is essential for understanding the function and behavior of cells under both healthy and diseased conditions.

Professors at Technical Institute of Physics and Chemistry, Chinese Academy of Sciences, have developed a new RNA dye - CP (emission wavelength of 658 nm, deep red fluorescence), which can be used for

  • monitoring the dynamic changes of nucleolar RNA in real time
  • completely recording the mitotic process of cells
  • evaluating the efficacy of anti-cancer drugs that targeted RNA polymerase I (Pol I)

Advantages

  • High selectivity and photostability
  • Rapid staining: rapidly stain nucleoli in living cells by binding with nucleolar RNA
  • Large Stokes shift: rigid-conjugated coumarin moiety is introduced into the molecular structure to increase the Stokes shift
  • Low cytotoxicity: can monitor nucleolar RNA changes during mitosis, and after treating with anti-cancer drugs cisplatin, actinomycin D and α-amanitin
